Screening of Polymorphic Microsatellite Loci in Lophophorus lhuysii by SPE and Its Application on Kinship Testing

Abstract:Chinese monals (Lophophorus lhuysii) are often socially housed in captivity with 2 females and 1 male in the unit. The exchange of males during the breeding season and the unfixed nest site for laying eggs have led to the vague recording of parentage information of L. lhuysii in the absence of genetic testing. To solve this problem, 13 microsatellite loci from the genome of L. lhuysii were isolated by routine method. In addition, the multiple amplification conditions of the primers were verified by SPE software. Specifically, the length of two multiple amplification PCR products differed by a single base was identified. The remaining 12 microsatellite loci were genotyped in 18 L. lhuysii eggshell/feather DNA samples. The sibling relationship between 15 individuals born between 2018-2019 was successfully completed. These markers can be used in the systematic control of kinship determination for L. lhuysii, and can also be used as powerful tools for the genetic management of L. lhuysii.
