Study on the Preparation and Control Effect of Egg Yolk Immunoglobulin of Anti-ApNPV

Abstract:Polyhedrosis of tussah silkworm caused by Antheraea pernyi nuclear polyhedrosis virus (ApNPV) is one of the main diseases in A. pernyi breeding. The yolk immunoglobulin is a rich sources of polyclonal antibodies, which provides cost, safe and efficient way to prevent and treat animal diseases. In this study, ApNPV was used to immunize egg-laying hens to prepare anti ApNPV immune yolk antibody (IgY). The IgY toxicity to A. pernyi was determined; and feeding tests were carried out to determine the optimum dilution ratio of IgY; the control effect of IgY was tested by feeding IgY in different manners. The results showed that: (1) the highest titer of IgY was 1∶210 at 6-8 weeks after immunization; (2) no adverse effects were found in the toxicity test, and the optimal dilution ratio of IgY was 64 times; (3) by feeding the 3rd instar larvae with IgY coated leaves prior to virus infection, the incidence rate decreased dramatically. It was also found that feeding the larvae with virus coated leaves first then followed by giving IgY coated leaves had positive effects. These results suggest that anti-ApNPV IgY has promising protective effects against infection by ApNPV, which may provide a potential strategy for control polyhedrosis of A. pernyi.
