COⅠ Sequence Analysis of Scirpophaga incertulas and its Application in DNA Barcoding

Abstract:Scirpophaga incertulas is one of the important pests affecting rice production. To investigate the genetic differentiation among different populations of S. incertulas in China, the mitochondrial DNA (mtDNA) cytochrome oxidase subunit (CO) of three geographic populations of S. incertulas was sequencedThe identity and variation of the sequences and their phylogenic relationships were analyzed by Mega 5.0. In this study, thirty six individuals of S. incertulas shared nine haplotypes. There were 17 polymorphic loci accounted for 3.13% of the total. The phylogenetic tree revealed no obvious geographical pattern for the prevalence and distribution of the haplotypes. Combining with the sequences of related species published online, genetic analyses were conducted and the results showed that CO sequence can be applied to species identification for S. incertulas.
